Regulation of Arachidonic Acid Release by Some Agonists on Suspended and Attached Neutrophils
نویسنده
چکیده
[H] Arachidonic acid is released after stimulation of suspended and attached neutrophils. The arachidonic acid release in attached cells is more than in suspended cells. This release is rapid with the increase of incubation time and dose depended. The Fmet-Leu-Phe (FMLP) potentiates [H] arachidonic acid release in attached cells pretreated with LPS and serum. [H] arachidonic acid release was less in suspended cells in comparison to attached cells where the addition of low concentration of LPS in presence of serum or platelet activating factor (PAF) to suspended cells produce increase in arachidonic acid release, where lps in combination with PAF produce a large and significant increase in arachidonic acid release. The potentiative effect of LPS is mediated via CD14 receptors. The monoclonal antibodies against CD14 had no effect on arachidonic acid release in PAF treated cells where it inhibited greatly the potentiation by LPS or LPS-serum complex with PAF.

Molecular mechanism of nucleotide-induced primary granule release in human neutrophils: role for the P2Y2 receptor.
Nucleotides are released during vascular injury from activated platelets and broken cells, which could stimulate human neutrophils. In this study, we characterized the P2Y receptors and investigated the functional effects of extracellular nucleotides on human neutrophils. Pharmacological characterization using selective agonists and pertussis toxin revealed that human neutrophils express only f...

[3H]Arachidonic acid is released after stimulation of rabbit neutrophils with fMet-Leu-Phe or platelet-activating factor (PAF). The release is rapid and dose-dependent, and is inhibited in phorbol 12-myristate 13-acetate (PMA)-treated rabbit neutrophils. The protein kinase C (PKC) inhibitor 1-(5-isoquinoline-sulphonyl)-2-methylpiperazine (H-7) prevents this inhibition. In addition, PMA increase...
متن کاملArachidonic acid-induced calcium influx in human platelets. Comparison with the effect of thrombin.
The effects of arachidonic acid and thrombin on calcium movements have been studied in fura-2-loaded platelets by a procedure which allows simultaneous monitoring of the uptake of manganese, a calcium surrogate for Ca2+ channels, and the release of Ca2+ from intracellular stores. Arachidonic acid induced both Ca2+ (Mn2+) entry through the plasma membrane and Ca2+ release from the intracellular ...
متن کاملInfluence of anti-inflammatory flavonoids on degranulation and arachidonic acid release in rat neutrophils.
We assessed the effects of 24 flavonoid derivatives, reported as anti-inflammatory, on lysosomal enzyme secretion and arachidonic acid release in rat neutrophils. Amentoflavone, quercetagetin-7-O-glucoside, apigenin, fisetin, kaempferol, luteolin and quercetin were the most potent inhibitors of beta-glucuronidase and lysozyme release. The first compound was also able to inhibit basal release. T...
